The crash occurred around 7 a.m. in the eastbound lanes of I-4 near the Orange-Osceola county line.
The FHP said a tire came off a vehicle and struck 47-year-old Ernie Centeno, of Kissimmee, on his motorcycle and led to the fatal crash.
Troopers said witnesses reported as Centeno was in the lanes of eastbound I-4, a gray pickup truck, described as a F-250 or F-350, ran over him and didn't stop.
Troopers are searching for the vehicle and any others that may have run over Centeno.
Traffic was backed up on I-4 for hours before being diverted off the highway. The interstate reopened just before 11 a.m.
The crash is still under investigation.
